Yeah SD is likely but not his only holding. Definitely charge him on the turn, he'll fold the river when he misses his draw which happens 4/5 of the time, whereas he'll pay a decent chunk on the turn very often.

what you reckon optimal bet size is here? like 1/3rd of the pot?

As much as they will call. It's a micro and you've less than pot behind, you could shove here frequently but it's ok to bet whatever and leave yourself a river gaybet if you think a shove is too much. Don't be surprised if they fold to the power 1/8th pot river shove either.

Generally at these stakes you should be trying to GII by the turn on drawy boards. The amount they are willing to pay to chase a draw drops dramatically on the river, can't think why.

Think if betting turn I would bet supersmall , so your opponent still has all 3 options left.

PS didnt see it was 50c maybe my comment is useless there.
