It depends. You want to iso opponents when it's advantageous to do so. This can involve various permutations of factors such as having cards that are stronger than his range, being more skilled post-flop, and having position, So when and how often you should look to iso people can be quite situational.

In NLHE its very common (eg: https://www.youtube.com/watch?v=5V3VhF3PSAk). As far as PLO goes I dont know much, but with so little preflop equity edges I dont think it has any sense to isolate (and its probably harder to do so since cold calling 3bets etc is very 'standard'). But maybe I'm wrong, I just started learning PLO :)

I think the problem with trying to iso in 5PLO is that you can't.

Too many people will call that iso rase or 3bet. And they won't be that wrong to do so.

The hand equities run pretty close, and people are too dumb to know what to fold. To be fair, most people don't play PLO that well. Which means everyone is a fish. Which means that playing tight, for value, in position is going to work wonders.

Don't worry about trying to isolate people until (1) it's worth it to isolate 1 bad player, (2) you have fold equity against the field, and (3) you have a solid reason to isolate 1 person v. all the other players at the table.
