PokerStars and 888poker are the largest rooms. You can always find there a lot of games.

Well i would say it depends on many things.. Which kind of poker do you prefer and at which time of the day you are playing most. Every poker room has it's "pro and contra"s.
Poker Stars has the largest player pool, which means the biggest MTTs and Cash Games with the most traffic in it. And it has one of the best poker software too.
888 has the best rewarding system in my opinion on the poker market, tons of Freerolls, Freebies, Daily "Wheel of Luck" and 888 Club. But the client can be looking a bit childish for many players and i hate it's table chat. It has slightly better rakes, than PS too.
Party Poker has also slightly better rakes, than PS, cashback system at 5$ rake per week or more and missions, which gives you additional bonuses. And of course Party Poker gives you the largest variety of online satellites to the big live poker tournaments around the Globe. If you are interested to play live for big money, but without investing much money for buy-ins, i definitely recommend to try this. It has also a solid software meanwhile too.
iPoker network rooms, such as Titan, William Hill and so on. Well it's known, that there are not so big player's pool in it, even though iPoker Network has the biggest amount of poker rooms in it and their games are merged (means you are playing in Titan poker with the players, who playing William Hill or NetBet or bet-at-home and so on), but it also known for it's weak field. (at least at Cash Games) Many poker coaches admitted, that you have usually better $/hour ratio playing there Cash, than at Poker Stars. They also have sometimes nice missions too. And Titan has some additional sweets for those who make rake, such as Gemstone Freerolls and Monthly Bonanza with $10,000 Prize Pool in it. William Hill also has some daily lotto-games for those who get to 2, 3 or 4 Level. Also they have SNG Ledder, where you can climb from just $1,1 to $500 winnings at the end. The only problem is the traffic, you should play at their prime time (Evening at Europe Time) in order to get any action.
ACR, BCP, Intertops and BetOnline - those are biggest American Rooms, i can't say much about contras, as i havn't had much experience playing there so far, but obvious Pro is that you have action if you are from Europe and prefer to play at Night, as it's a Day in America and the largest traffic in there, while at other poker rooms, have been listed above, really just a few action. Also i've heard about it's weaker field of players. (a lot of fish)
Hope it helps
